On the ground, you should see a beetle (either brown or regular). From the bottom of the stairs, take a right and go to the water tanks. Shoot it to make it drop, then head back down the stairs to collect it. On the wall above the tunnel that you should be facing, you'll see a ruby. In Chapter 5-1, climb to the stairs that are positioned directly ahead of you as you start the stage.
